Thermostable ?-amylases are being used in a wide range industries, including food, textiles, detergents, pharmaceuticals, fine chemicals. A good source thermostable thermophilic is found high-temperature habitats like hot springs. Hence, this study aimed to screen, isolate, characterize (APB) the spring Sabah. Sediment water samples were collected springs, serially diluted, plated onto Luria Bertani agar medium containing starch, incubated at 60°C for 48 hours. The bacterium was identified by halo formation around colony after stained with Lugol's solution. Nine colonies be able form zones, creamy (A7 strain) producing highest amylolytic index (4.24). Further A7 strain showed that isolate Gram-positive, rod-shaped bacterium, positive reaction upon oxidase catalase tests. 16S rRNA sequence had 99.81% similarity Anoxybacillus flavithermus, therefore as A. flavithermus strain. Further, growth curve analysis indicated grew well 60°C. 3, 5-dinitrosalicyclic acid (DNS) assay crude enzyme secreted exhibited optimum amylase activity 8.6 x 10-2 U/ml. This first APB obtained springs Sabah promising further studies applications.
